Investigation of association between coagulation factor v, factor II, methylenetetrahydrofolate reductase, tumor necrosis factor alpha and plasminogen activator inhibitor genetic variations and stroke: a case-control study in an Iranian population with high incidence rate of stroke

چکیده
Introduction: Stroke is a multifactorial disorder caused by a combination of genetic and environmental riskfactors. The incidence of stroke in Iran is considerably greater than in most Western countries with strokeoccurring at younger ages. Many studies provide substantial evidences for a genetic contribution to strokedisease. This gene association study was aimed to determine the association between factor v, factor II,methylenetetrahydrofolate reductase, tumor necrosis factor alpha and plasminogen activator inhibitor geneticvariations and stroke in an Iranian population.Material and methods: A hundred and fourteen case and 114 control enrolled in this case control study.Matched case and control subjects were randomly selected from same geographically area. Genetic variant of FVc.1691 G> A, FII g.20210G> A, MTFHR c.677 C> T, MTFHR c.1298 A> C were investigated by FRET real timePCR assay while the genetic variation of TNF-a -308 G> A and PAI-I -675 4G> 5G were determined by RFLPPCR.Results: there was no correlation between factor V leiden c.1691 G> A, FII g.20210 G> A and MTFHR c. 1298A> C variations and stroke. There was a significant difference between TNF-a -308 G> A, PAI-I -675 4G> 5G andMTFHR c.677 C> T genotype distribution and stroke.Conclusion: TNF-a -308 G> A, PAI-I -675 4G> 5G and MTFHR c.677 C> T genotype variations could beconsidered as candidate genetic risk factors for high incidence rate of stroke in north east of Iran.کلیدواژه ها
