Significant attenuation of specific activity of cytochrome C oxidase in brain of mouse model of Multiple Sclerosis disease

چکیده

Introduction: Multiple sclerosis (MS) is a chronic, debilitating disease characterized by the invasion of theimmune system to the central nervous system components including brain, spinal cord and optic nerve.Deficiency in mitochondrial energy supplement also affect the complete myelination, followed by inflammationin neurons occurred in patients with MS. The aim of this study is evaluation of specific activity of cytochrome Coxidase (COX) in brain tissue of mouse model of MS disease (EAE) which is an important enzyme in electrontransferchain in mitochondria to produce ATP in brain.Methods: The C57BL/6 mice were purchased from the Pasture Institute and Hooke kit used for inducing EAE.High Score of this model (3 to 4) used in this study. The brain tissues of mice were examined for determining oftotal protein and activity of COX in 3 group of sham, negative control and EAE mice.Results: Specific activity of cytochrome C oxidase was significantly decreased (more than 3 fold) in EAE modelin comparison with sham and negative control. Meanwhile, there are no significant differences between negativecontrol and sham.Conclusion: Significant attenuation of specific activity of COX in EAE model indicated that the function ofmitochondria of the nervous system of EAE impaired to produce of energy affecting of neurogenerative processin this model of MS disease.
