Sorting nexin expression involved in endosomal trafficking is decreased in high EDSS of multiple sclerosis

چکیده

Introduction: Multiple sclerosis (MS) is a chronic inflammatory and demyelinating disorder of central nervoussystem (CNS). MS is characterized by the damage of myelin, chronic inflammation, axonal and oligodendrocytesinjury and progressive neurologic disorder. Retromers is a protein complex that has an essential role in endosomaltrafficking, and retromer dysfunction has been associated with a numerous of neurological disorders. Therefore,this study aimed to compare the expression of sorting nexin2 gene as a part of retromer complex in MS patientswith health control and evaluation correlation between gene expression and disability level (EDSS).Methods: In this case-control study, 100 individual (50 patients with MS and 50 controls) were enrolled.Followed verifying disease and sampling, total RNA was extracted and complementary DNA (cDNA) wassynthesized. Patient’s disabilities were assessed by Kurtzke Expanded Disability Status Scale (EDSS). The relativegene expression evaluated by QPCR and data analyzed.Results: The findings demonstrated a significant association between the sorting nexin expression and MS(P=0.01). Moreover, a decreased expression was observed in patients with moderate and severe disability (EDSS6-10) and it was statistically significant (P < 0.05).Conclusion: this study shown that expression of sorting nexin2 is lower in MS patient and may be affectedinability of individual. Considering the functional role of sorting nexin2 as a protein involved in traffickingprocess, may affect receptor function or drug targeting.
