Introduction & Aim: Metastasis is the leading reason for the resultant mortality of patients with cancer. The past few decades’ remarkable progress has been made in understanding the molecular and cellular basis of metastasis in cancer. High expression of Stromal-derived factor-1 (SDF-1), an alpha-chemokine that binds to G-protein-coupled CXCR4, is involve in invasiveness and migration of breast cancer cells metastasis to organs (bone, liver, and lung). The SDF-1 levels correlated with node involvement and long-term survival in patients with breast cancer therefore it may potential value in assessing clinical outcomes of patients with breast cancer. In the proposed study using the breast cancer tissues from patients showed the active SDF-1/CXCR4 pathway, are more invasive and motile, thus have a more aggressive phenotype. The results will provide an in-depth understanding of SDF-1/CXCR4 pathway correlated with nodal status, recurrence and, most notably, both overall and disease survival indicates that SDF-1 and indeed the SDF-1 receptor complex has strong predictive value in assessing long-term clinical outcomeMethods: Tissue samples were collected from patients with invasive ductal carcinoma from Kerman (n=24) and Tabriz (n=23). Samples from health tissues used as controls (n=7). The cDNA was synthesized from RNA, the expression of SDF-1 and CXCR-4 genes were analyzed by RT-PCR data were analyzed with ∆∆CT method. Results and Conclusion: We detected high level of chemokine SDF-1 and its receptor CXCR4 mRNAs in invasive ductal cancer samples of both patient groups from Kerman and Tabriz. The present study represents a significant step forward to understand the SDF-1/CXCR4 level in tumor tissues correlated with node involvement and we suggest that diagnostic tools and therapies targeting which can improve the clinical outcome.
