In vitro antitumor effect of metformin in murine 4T1 breast cancer cell line

Introduction: Breast cancer is the most commonly diagnosed cancer in women. Previous studies have showed that the anti-diabetic drug metformin as potential anti-tumor agent that could targeting breast cancer cell. Thus this study was designed to evaluate the anti-tumoral effect of metformin on 4T1 breast cancer cell line. Methods: 4T1 cells were cultured in RPMI supplemented 10% FBS and incubated in 5% co2 at 37°C. Then the effect of metformin in the different concentrations (0-10-20-40-60-80-100 m mol/l) was tested by MTT assay and the cell viability was measured after 5 day incubation. Results: The result of MTT assay showed that the IC50 value (i.e., the concentration of metformin required to decrease MTT metabolization by 50%) ranged from 60±2 m mol/l metformin in 4T1 breast canc er cell Conclusion: These results demonstrated that metformin could be used an effective antitumor drug, and has the potential to developed as a new anticancer drug
