Galectin-9 induces cell cycle arrest in breast cancer cells through inhibition of cyclin D1/CDK-4

چکیده

Introduction and aim: Dysregulation of cell cycle is a major hallmark in cancer biology that might equip tumors with a higher malignant potential and chemoresistance. Galectin-9 that modulates a variety of biological functions including proliferation, as well as apoptosis of tumor cells. In this study we evaluated the role of Galectin-9 in cell cycle progression in breast cancer cells. Material methods: Cell viability was measured by MTT assay in MCF-7 and MDA-MB-231 breast cancer cell line. Analysis of cell cycle distribution carried out by flow cytometry. To determine the mechanisms of Galectin-9 -mediated induction of cell cycle arrest, the expression of cell cycle regulatory proteins cyclin D1 and CDK4 were evaluated. Result: Our result showed that Galectin-9 significantly reduced Cell viability in a dose-dependent manner.Moreover, our results indicated that Galectin-9 induced G1 cell cycle arrest. Reduction of cyclin D1 and Cdk4 protein levels was also observed after treatment cancer cells with Galectin-9.Conclusion: In conclusions this study demonstrated that Galectin-9 Induces G1 Phase Cell-Cycle arrest through cyclinD1/ Cdk4 mediated pathway in ovarian Cancer Cells
