The study of anti-inflammatory and anti-oxidant effect of l-carnitine in mice with induced polycystic ovary syndrome

Background: Hyperandrogenism, anovulation and insulin resistance are symptoms of polycystic ovary syndrome (PCOS) related to low levels of serum L-carnitine. Inflammation and oxidative stress are involved in the pathogenesis of PCOS. The aim of this study was to investigate the effects of L-carnitine as an anti-oxidant in mice with experimental PCOS.Methods: PCOS was induced through daily injections of testosterone enanthate (1 mg/100g body weight s.c.) for a period of 5 weeks. NMRI mice (3 weeks old) were divided into 4 groups (n=6): Control, PCOS, PCOS + L-carnitine and L-carnitine.Treatment was carried out with the dose of 500 mg/kg by i.p injections every other day for 4 weeks. At the end of treatment, serum levels of IL-6 and TNF-α, Malondialdehyde (MDA) and the antioxidant capacity were measured relatively using the ELISA kit, thiobarbituric acid (TBA) and Ferric reducing antioxidant power (FRAP) assay. Data were analyzed using one way ANOVA and Tukey s test and the means were considered significantly different at pResult: The serum levels of IL-6, TNF-α and MDA significantly increased in the PCOS group compared to the control one while these parameters significantly reduced to the control level in the PCOS +L-carnitine group. Antioxidant capacity also reduced significantly in the PCOS group when compared to the control while it significantly increased in the PCOS+L-carnitine group to the control level.Conclusion: L-carnitine could prevent oxidative stress and ameliorate the inflammatory markers in mice with PCOS.
