Investigating callus induction from seed coat layer of quince for mucilage production

چکیده

Mucilages are abundant in nature and present in many plant sources. Quince (Cydonia oblonga Miller) is a tree from Rosaceae family. Different parts of quince have been used as traditional remedies. The seeds are used medicinally, and have high value as demulcent tonic. Mucilage is derived from the cells of the epidermis of the seed-coat. The mucilage is a complex of cellulosic fraction with a more readily hydrolyzed polysaccharide. The aim of the present investigation was to study mucilage content of callus inducted from seed coat layer of quince. This seed coat layers were cultured on Murashige and Skoog medium supplemented with different concentrations and combinations of NAA (0.2mg/l) and 2,4-D (0,5,10,20 mg/l) for callus induction and incubated at a temperature of 25±1°C for 4 week in darkness then were transferred to incubator with a 16 to 8 light/dark cycle. The highest frequency of callus production (17.9%) was observed in the medium consisting of 2,4-D (20 mg/l) + NAA(0.2 mg/l) and maximum quantative amount of callus (0.31) was obtained in the medium containing 2,4-D (5 mg/l) + NAA (0.2 mg/l). The results from HPLC analysis of the callus s mucilage will be presented later.
