Making curcumin-albumin nano-drug and evaluation of its properties on sk-br3 (breast cancer her2+ cell line)

Today, research on the use of Nano compounds in the cancer treatment has become so common. The clinical use of many plant antioxidants is limited by the inability to dissolve in water and biologicalfluids, so researchers hope that nanotechnology can overcome these problems. Among herbal antioxidants, curcumin is one of the strongest antioxidants to inhibit the proliferation of cancer cells whose clinical use is limited due to the very low solubility in water and instability in physiological conditions, especially around the tumor. To overcome these problems, various compounds of Nano- curcumin have been produced and tested. The important thing is that the Nano substance must be harmless, protect the anti-cancer properties of curcumin and also increase its solubility in water and its stability in the blood. In this study, curcumin was had conjugated with albumin, a completely safe bio compound, and was had evaluated its properties.Briefly, the solution of curcumin in ethanol has been mixed with albumin, then stirred for 24 hours. Subsequently, the precipitate was washed 5 times, and the bath sonicator was performed. Finally, lyophilization was performed. The resulting powder was tested by FTIR, DLS, Zeta potential, TEM and SEM. The amount of its load and release was measured in normal and tumor conditions. In the end, this combination has tested on the SKBR3 cell line to examine its anticancer properties.The FTIR confirmed that curcumin and albumin bonded together. Particle size was reported 220 and 180 nm by DLS and SEM, respectively. The zeta potential of the Curcumin-Albumin compound was -7 while curcumin zeta potential was -37. So zeta potential of this compound was 30 V more positive than curcumin. TEM showed that each albumin is bound to 8 to 10 particles of curcumin. The drug loading was about 40%, and the release of the drug lasted 7 days in normal condition and 5 days in tumor condition. MTT tests indicated that the toxicity of this drug on the normal cells is less than curcumin, but its anti-cancer property on the SKBR3 cell line is much greater than curcumin .findings of this study showed that curcumin-albumin compound not only had anti-cancer property of curcumin but caused decreased curcumin toxicity on normal cells and increased its anti-cancer property.(by Controlled of curcumin release around of cells both normal and tumor)
