Nanoparticles have a potential use in wide range of fields including medicine. Metal nanoparticles havea variety of applications in cancer treatment. The zinc oxide (Zn O) nanoparticles have acceptableeffects for cancer treatment. The purpose of this study is to investigate the cytotoxicity of zinc oxide nanoparticles synthesized in plant Ferula assafoetida extract on HT-29 cells (colon adenocarcinoma cellline). Ferula assafoetida is a native plant of Iran that has many remedial properties.HT-29 cells werecultured in a laboratory and treated with different concentrations of the nanoparticle and then tested using MTT assay. The cells were seeded in 96-well plates and treated for 24, 48, 72 hours. IC50 values calculated according to MTT method. cytotoxicity of Zn O nanoparticles on HT-29 cells showed thatnanoparticles were able to inhibit cancer cells with IC50 values of 50, 125 and 125 μg/ml at 24, 48 and 72 hour-time intervals, respectively. The results showed that time and concentration are effectivefactors in nanoparticle cytotoxicity. With the increasing of the concentration from 31/2 to 500 μg/ml in 48 hour-time interval cell viability decreases from 92% to 31%. The effect of time on nanoparticletoxicity showed that there was a significant decrease in the amount of cell viability from 24 hours to 48 hours. However, the values of IC50 did not change significantly from 48 to 72 hour-time intervals.Results of this study indicated that inhibition effects of Zn O nanoparticles synthesized through green chemistry, in the growth of colon cancer cells were observed in this experiment. According to theresults, this nanoparticle can be used in further testing in order to use in the clinic
