Study of genetic diversity among (Phaseolous vulgaris) genotypes using molecular markers

The genetic structure of 21 common bean lines collected from different regions of Iran were evaluated using 12 different combinations of ISSR markers having 94 polymorphic bands. Among the studied markers, UBC917marker with 14 polymorphic bands, and UBC823marker, with 11 polymorphic bands, produced highest and lowest polymorphic bands, respectively. In addition, UBC823and UBC815markers were 100% polymorph and generated 8 and 10 bands, respectively. Estimation of the number of observed alleles and number of effective alleles ranged between 1.7 to 1.82 (for markers UBC825and UBC816) respectively. Calculation of Nei gene diversity and Shannon index showed that UBC813and UBC814markers had the lowest Nei gene diversity and Shannon index (with 0.4 to 0.59), while the highest Nei genetic diversity and the Shannon index were observed for UBC812marker. Polymorphism information content (PIC) of markers was ranged between 0.34 to 0.48, and the effective multiple ratio (EMR) was ranged between 4.9 to 8.10. Marker index estimation ranged between 2.35 to 4.83, for UBC812and UBC824markers, respectively. Principal coordinates analysis showed that the first two components, explained a total of 20.01% of the total variation, which illustrates the effective use of these markers and their distribution in the genome of common been for estimating the variation between populations. Cluster analysis using UPGMA method and simple matching coefficient divided the 21 populations into 4 main groups which included 1, 6,12 and 2 individuals, respectively.
