Chitosan is a biodegradable natural polymer that it is soluble only in acidic environment and this restricts its applications. So carboxymethyl chitosan (CMC) that is water-soluble chitosan by-products can be applied for biomedical applications such as preparation biodegradable nanoparticles for drug delivery. In this research chitosan derivative (CMC) was synthesized and CMC nanoparticles were prepared by ionic gelation with calcium ions using calcium chloride (CaCl2). The objective of this study was to preparing of CMC/ CaCl2 nanoparticles with different particle size and spherical shape for using in drug delivery systems. The factors affecting nanoparticle formation were studied and have been characterized. The effect of various parameters on the formation of CMC/ CaCl2 nanoparticles investigated and found that particle size was highly influenced by CMC concentration, CaCl2 concentration, and mass ratio of CMC to CaCl2. As it observed, with increasing CMC concentration the particle size and standard deviation (SD) of CMC nanoparticles were increased and poly disparity index (PDI) was decreased
