چکیده مقاله:

In spite of advanced treatment, cancer, the second cause of death in the world, is expanding. Malignant melanoma considered between ten common cancers globally, has been associated high mortality statistics to itself. Flavonoids, polyphenol compound, which are rich in plants and vegetables, developed more demands. Myricetin because of its structure (the hydroxyl groups and C=C bound), took priority over other flavonoid subclasses. In this study, we investigated the anti- proliferative effect of myricetin on melanoma cells, so A375 cells were treated with different concentration of myricetin (0 to 100 µΜ) in three period of times (24, 48, and 72 h). The cell viability was determined by MTT (Methylthiazolyl diphenyl-tetrazolium bromide) assay. After considered times, the MTT solution was added, then the viability of cells were detected by measuring the absorbance on 570nm relative to 630 nm. Our finding showed that low concentration of myricetin (up to 25µM) has no toxicity effect. Also the result confirmed the IC50 of myricetin on melanoma cells for three ordered period (24, 48, 72 hour) as following: 50, 40, 35µΜ, respectively. According to this research, myricetin has anti-proliferative effect on melanoma cells which can be used as a therapeutic agent accompanied by other treatments and this study could be a breakthrough in future researches to acquire confirmative results
