The Correlation Analysis of H. Pylori Infection on Cellular Stress Pathway Expression in Gastric Cancer

چکیده

Identification of cellular stress pathways can be offered a possible therapeutic procedure forgastric cancer (GC) using activation of the apoptotic response pathways. GRP94 and Chopare involved in the process of survival and apoptosis respectively, dependent on theendoplasmic reticulum (ER) stress as an unfolded protein response (UPR) pathway. Thepresence of Helicobacter pylori (H. pylori), the oncogenic factors production as well assubsequently developing inflammation are considered as efficient factors in the GC. The aimof this study was to investigate the role GRP94 and Chop in the progression of GC and theircorrelation with clinicopathological features in GC patients. In addition, we examined gastrictissue samples for H. pylori DNA. This study was assessed GRP94 and Chop mRNAexpression using quantitative real-time PCR from fresh tumoral tissues and distant tumor-freesamples of 50 GC patients. The presence of H. pylori infection was also detected using UreCand 16S rRNA genes by PCR. Of the 50 samples, 22 (44%) cases were confirmed for thepresence of H. pylori by PCR. GRP94/Chop overexpression was detected in 8 and 17 of the50 (34 and 16%) GC samples respectively. There was a significant correlation betweenexpression levels of GRP94 with Chop (p value< 0.0001). Moreover, a significant associationwas also detected between overexpression of Chop with gender (p= 0.048) and tumorinvasion (p= 0.029). We demonstrate that human GC cells produce Chop and GRP94 and asignificant correlation was seen between Chop overexpression and GC tumor invasion,although Chop overexpression cannot induce cell apoptosis in GC cells.
