چکیده مقاله:
A wide range of microorganisms and microarthropods are known as polluters of plant tissue culture. The outcomes of pollution in tissue culture include increase in costs, negative effectson cultures, false test results, and loss of valuable materials. Successful usage of Silver Nanoparticles (AgNPs) in different medical methods as antifungal and antibacterial reagentsled to their application in controlling plant pathogens. With a wide range of antimicrobial activities, Silver Nanoparticles decrease different Plant diseases which are caused by spores that produce fungal pathogens. This research evaluated microbial pollution of Kiwi tissueculture by use of Silver Nanoparticles. this experiment was conducted in a completely randomized design within 16 groups, each of which including 5 explants. Lateral buds were disinfected in four different concentration of Nano-Silver (25, 50, 100,200 mg/L) in differenttimes, and then, compared to control samples which were disinfected by sodium hypochlorite and sodium hypochlorite with mercuric chloride. Explants were cultured in a MS culture environment which was enriched by 2 mg/L of BA hormone. Finally, to evaluate the effectsof Nano-Silver on quantitative and qualitative characteristics of cultured explants, they were statistically analyzed. Application of Nano-Silver in different concentrations and appointed time intervals led to different results. Treatment of pieces when Nano-Silver was not appliedhad the most pollution in control sample A. Also the most amount of pollution was seen for concentration of 25 mg/L, and time interval of 15 minutes when Nano-Silver was had been applied. The least amount of pollution when Nano-Silver had not been applied was related to control sample B. Finally, the treatment of pieces in concentration of 100mg/L and time interval of 180 minutes led to omission of all bacterial and fungal pollutions. The results of the TEM electron microscopy shows the presence of silver nanoparticles kiwi plants.
