Protein Immobilization on Carbon Nanotubes as a Template for Drug Delivery Applications

Through the organic functionalisation of carbon nanotubes, the prerequisites for their solubility profiles and their manipulation into biological systems are established. In particular, these fuctionalized carbon naotubes can be further bonded to bioactive peptides, proteins and nucleic acids to attain biocompatibility. Immobilisation of proteins to the external walls of carbon nanotubes may be find interesting applications in diagnostics, vaccine and drug delivery, or multipresentation of bioactive molecules. In this study, we report first the functionalisation of CNTs using strong acids, resulting in the generation of carboxylic groups, which increase their dispersibility in aqueous solutions and facilitate their reaction with the amino groups present in the biological molecules. In the next step the covalent bonding of globular protein bovine serum albumin (BSA) to multi-walled carbon naotubes (CNx MWNT) via a two-step process of diimide –activated amidation was investigated. Subsequently CNT-BSA conjugate with fluorescein isothiocyanate (FITC) was derivatised. The FITC-CNT-BSA will facilitate further examination of internalization of these conjugates into cells. The former was then confirmed via FTIR measurements, and the latter was proven by AFM results which successfully indicate the attachment between the protein and CNx MWNT.
