Intraperitoneal Immunization with Recombinant Urease Loaded N-trimethyl Chitosan Nanoparticles Confers Protection against Brucella abortus and Brucella melitensis in Mice

چکیده مقاله:

Brucellosis is a world prevalent endemic illness that is transmitted from domestic animals to humans.Infectious bacteria use urease activity in various ways along pathogenesis. Brucella spp exploit urease enzyme forsurvival in the harsh conditions of stomach during the gastrointestinal infection. In this study, we examined the immuneresponse and the protection elicited by using recombinant Brucella urease Loaded N-trimethyl Chitosan (TMC/Urease)nanoparticles in BALB/c mice. Recombinant urease with Freund’s adjuvant (Urease-IFA) was administeredintraperitoneally while TMC/Urease nanoparticles was administered orally and intraperitoneally. After in vitrostimulation, splenocytes from intraperitoneally vaccinated mice with Urease-IFA and TMC/Urease nanoparticlesproduced IL-4, IL-12 and IFN-γ, while orally vaccinated mice produced IL-12, IFN-γ and IL-17 titers. Immunized micewere challenged with virulent B. melitensis and B. abortus. Intraperitoneal immunization with TMC/Urease nanoparticlesexhibited the high protection level against B. melitensis and B. abortus. Altogether, our results indicated that TMC/Ureasenanovaccine could be a useful vaccine candidate against brucellosis.
