Metabolomic approaches are used for understanding regulatory networks and metabolite fingerprinting involved in the plant kingdom. Because of the chemical diversity of cellular metabolites, there is no single technique suitable for measurement of all metabolites. In the most of analytical methods, gas chromatography (GC)/mass spectrometry (MS) system has been developed for detection of secondary metabolite profiling. In this investigation, we described a rapid GC/MS-based metabolite profiling method in damask rose. In this study, a detached flower petal of damask rose is inserted in a glass vial which is closed with a PTFE-lined septum and cap. The production of volatiles by the petal continues for several hours, and all volatiles are accumulated in the headspace of the petal. The vial can be placed in the sampling tray of a GC/MS system with a Combi PAL robotic injector. The main secondary metabolites identified in Rosa damascena were as follows: phenyl ethyl alcohol, β- citronellol, α-pinene and geranyl acetate. In this way, it is possible to screen the patterns of emitted secondary metaboliteprofiling of a large number of plant samples at high-throughput level. This method would be a great theoretical impact on understanding metabolism, for example for metabolite fingerprinting and the prediction of novel metabolic pathways.
