چکیده مقاله:
DNA polymerases are a family of enzymes responsiblefor replication,maintenance and transmission ofgenetic information in all organisms. Both prokaryotesand eucaryotes have several DNA polymerases ; anumber of them are involved in DNA replication butothers are responsible for repair and other accessoryprocesses. DNA polymerase have been used extensivelyin molecular biological, specially in two importanttechnologies in molecular biology: PCR and dideoxyDNA cycle sequencing.Since the discovery of the first thermostable DNApolymerase –Taq polymerase - and its applicationin PCR ,molecular biology research has beenconsiderably expedited and search has been directedfor other thermophilic DNA polymerases with morerefined qualities.The aim of the present study was toassess the potential biotechnological capabilities ofIranian thermostable DNA polymerases from variousthermophilic eubacteria isolated from a hot spring inArdabil, Iran. Biochemical and microscopic analysisshowed that they belong to the genus Bacillus andGeobacillus. Appropriate primers from highly conserveddomains of this gene in thermophilic Bacillus weredesigned. From the nine isolates examined , five wereselected for further characterization of the gene.
