The effect of cryopreservation on germination and growth indices in some orthodox seed of medicinal and rangeland plants

چکیده مقاله:
With regard to growing process of natural overthrowingand germplasm resources destroying in our country,as one of the most rich fullness countries on plantgenetic resources, it is necessary to act fundamentallyon conservation and maintenance of available geneticdiversity. By using seed cryopreservation, as one ofthe Ex situ plant germplast conservation method withunlimited seed storing without losing seed viability, wecan provide a seed conservation method that its costis %1 of In situ plant germplasm conservation. In thisstudy the effect of cryopreservation on germinationand growth indices (germination percent, germinationrate, seed vigour index, plantlet length, plantlet freshweight) on orthodox seed of seven medicinal andrangeland plant species (Artemisia khorasanica,Hypericum perforatum, Origanum spp, Trigonellamonthana, Phytolaca spp., Hypericum anderosaemum,Daturea spp.) were evaluated. Results showed thatthere is significantly difference between plant specieson all evaluated traits that it is because of differentpotentially genetic diversity of these plants. Moreover, incomparison of cryopreservation and control treatmentsin each species, no significant different was observedexcept for germination percent and germination rate. Itis obvious as orthodox seeds are tolerant to desiccationand stay in inactive status on cryopreservation. We canemphasis cryopreservation as a suitable, alternativeand inexpensive method for healthy long term storageof orthodox seed in germplasm resources conservationcenters
