In this study, a novel and sensitive UV-Vis spectrophotometric method was developed for the determination of clindamycin based on its interaction with a mixed-ligand system composed of Bromopyrogallol Red (BPR) and Methylene Blue (MB). The complex formation was monitored at an absorption maximum of ۵۱۰ nm, showing a pronounced increase in absorbance with increasing clindamycin concentration. Various spectroscopic studies, including Job's method, the molar ratio method, and Benesi-Hildebrand analysis, confirmed a ۱:۱ stoichiometric interaction between clindamycin and the ligand system. The effect of pH, temperature, and reaction time was thoroughly optimized, with the best results achieved at pH ۴.۵ using an acetate buffer. Interference studies demonstrated the method's high selectivity against common ions, excipients, and structurally related drugs. The developed method was successfully applied to pharmaceutical formulations and spiked biological samples (serum and urine), with excellent recovery values (۹۴-۱۰۲%) and low relative standard deviations. The method is rapid, cost-effective, and suitable for routine analysis in quality control and clinical laboratories.
