Hyaluronic Acid/Chitosan Hydrogel: Biomimetic Wound Dressing Inspired by the Inherent Structure of the Skin on Diabetic Wounds

چکیده

Background and purpose: According to the statistics of the Health Organization (۲۰۲۴), diabetic foot ulcer (DFU) includes a high rate of chronic disorders, which has an increasing trend of complications of limb amputation and death, and will have a high economic effect on the health of society. So far, there have been many studies on dressings, now we have designed a hydrogel inspired by the inherent properties of the skin with a rich source of chitosan and hyaluronic acid, which can play an effective role in better tissue regeneration and the extracellular matrix of wounded skin. Materials and methods: In this study, we used chitosan/hyaluronic acid/hydroxypropyl methyl cellulose scaffold network to preparation hydrogel. Also, for better drug delivery, we used silver nanoparticle by lipid transfer system. For animal research, ۸ male Wistar rats of ۲۰۰ grams were used by DFU model with ۱۲ mm diameter, which were divided into two subgroups (n=۴) and evaluated of wound closure rate, SOD enzyme concentration were performed at different times, as well as antibacterial evaluations using the aura of non-growth and non-toxicity was performed on human skin fibroblast cells. In order to check the inherent properties of the hydrogel, water solubility, water vapor transfer, swelling, degradation tests were performed. Results: The results showed that the release rate of silver nanoparticles in the hydrogel was continuous and controlled delivery. In the animal evaluation of the closure of the wound surface, a ۹۵% reduction in the level of damage was observed. In the antioxidant evaluation with the DPPH technique, due to the presence of natural polysaccharides, was observed a decrease in reactive oxygen species, and the antioxidant enzyme concentration had a significant level compared to the control group. In the antibacterial evaluation, a non-growth halo was observed, in the cytotoxicity test by the MTT hydrogel method, ۱۲۶% cell viability and non-toxicity were observed after ۴۸ hours on human skin fibroblast cells. The swelling property of the hydrogel was reported to be ۴۴%. A water vapor transmission rate of ۲۰۲۸ was reported to provide an ideal environment for secretion absorption and cell migration.
