Targeted Treatment Based on Biological Nanoliposome Carrying Collagen and Recombinant Complex: A New Therapeutic Strategy in Wound Healing

چکیده

Background: Bio-nanoliposome carrying fast-absorbing collagen and recombination complex is an important biopolymer with a three-dimensional structure produced by modern nanotechnology and has been widely used in medicine. One of the unique characteristics of this nanoliposome is its very high-water absorption, which can be used to heal deep and extensive wounds; But after drying, its three-dimensional structure collapses and its water absorption decreases. This research aims to preserve the three-dimensional structure of nanoliposomes with the help of their cross-linking and to improve the water reabsorption properties of this biopolymer. In addition, the survival, proliferation, and cell growth rate of the nanoliposome structure carrying these modified and untreated strategic materials were also studied. Materials and methods: In this research, nanoliposome was produced, purified, and neutralized using static culture. Inside this nanocarrier, tissue-absorbing collagen, and tissue-restoring complexes such as minerals, vitamins, and Flaxman recombinant protein were loaded; The design system of nanoliposome surface spikes was based on the Advent Pro method. Then the samples were treated with different concentrations of citric acid/sodium hypophosphite and networked. Finally, the characteristics of processed and raw samples were checked with various tests, including ATR-FTIR, MTT, SEM, water absorption, and in vitro and in vivo tests. ۱۰۰ mice divided into four groups, ۲۵ mice in the control group, ۲۵ mice receiving nanoliposomes designed to include grade ۳ wounds in the femoral area of the hand, ۲۵ mice receiving nanoliposomes designed to include grade ۳ wounds in the italic area of the foot, and ۲۵ mice with type ۲ diabetic compound wounds in the back area. They were the backbone. Routine serology and blood tests, as well as routine liver enzymes (ALT, AST, ALP) along with an HR-CT scan two days in between with contrast, were performed and the laboratory results of recombinant protein quality tests were performed using ELISA and HPLC. Results: Based on the obtained results, it was found that the cross-linking operation prevents the structure from collapsing and not only does not cause toxicity, but in addition to increasing the amount of water absorption, it also increases the viability, adhesion, and proliferation of cells in the repaired
