Characterization of stem cells isolated from peritoneal dialysed fluid

چکیده
The mesothelium, composed of monolayer of mesothelial cells (MCs) attached on a thin base membrane supported by a subserosal connective tissue, suggested as a dynamic structure that plays an important role in homeostasis, wound healing, fluid transport and inflammation. The introduction of peritoneal dialysis (PD) as a modality of renal replacement therapy has provoked much interest in the biology of the peritoneal MC. In the present study, we identified cells that express some defined markers of mesenchymal, hematopoietic and embryonic stem cells in human peritoneal mesothelial cells ( HPMCs).Materials and methods: In order to investigate the nature and plasticity of MCs isolated from peritoneal dialyzed fluid (PDF), cell populations from PDF of early stage non-peritonitis patients were screened for evaluating of dominant marker determination by cell culture, immunocytochemical, immunofluorescence and RT-PCR. Isolated cells were cultured and then in the first and 4th passages were studied for cytokeratine 18, HBME-1, CD34, CD38, CD90, CD452and alkaline phosphatase (ALP). Specific media for differentiation mesothelial cells to osteoblast cells were difined. Results: Our results were positive for some of defined markers and negative for the others. The formation of osteoblast like cells from mesothelial cells in specific media was confirmed by expression of osteopontin and ALP genes and alizarin red staining. Conclusion: It seems peritoneal MCs in culture possess immunocytochemical markers identical to mesothelial stem cells. This study suggests a multipotent plasticity of mesothelial cells.کلیدواژه ها
