Anticonvulsant potentiating effect of ferulic acid on sodium valproate and carbamazepine in mice maximal electroshock seizure model: a mechanistic perspective

Ferulic acid (FA) is a plant phenolic acid with antioxidant, anti-inflammatory, and neuroprotective effects. The present study investigated the effect of FA alone and combined with sodium valproate (VPA) or carbamazepine (CBZ) on seizure severity, and associated changes in brain interleukin-۱β and antioxidant reserve of mice after maximal electroshock seizure (MES) induction. Male mice (weight: ۲۵ ± ۵ g) were used in this study. Animals were treated with saline; FA (۲۰ and ۸۰ mg/kg); diazepam (D) (۲۰ mg/kg); FA (۲۰ mg/kg) + VPA (۲۰۰ mg/kg); FA (۲۰ mg/kg) + CBZ (۱۰ mg/kg) for ۷ days before MES. The duration of hind limb extension (HLE) were recorded. Mice motor coordination, as a side-effect of treatments, was investigated with chimney test. In addition, interleukin-۱beta (IL-۱β), and total antioxidant capacity (TAC) levels in the brain were evaluated. The administration of FA in both doses significantly decreased the HLE duration compared to the saline treated group. FA (۲۰ mg/kg, ip) was significantly increased the anticonvulsant effect of VPA and CBZ. Both doses of FA protected from seizure-induced TAC decrease and of IL-۱β increase in the brain of seizure mice. FA (۲۰ mg/kg, ip) + VPA treatment showed more protective effect in mice against brain TAC reduction and increase of IL-۱β levels than each one. But FA (۲۰ mg/kg, ip) + CBZ had no synergistic effect on both measured parameters. All treatments had no significant effect on the motor coordination in mice. The results of the present study showed the neuroprotective effect of the FA and its enchaining effect on VPA and CBZ against MES. More effective attenuation of seizures, neuroinflammation and oxidative stress by FA + VPA co-treatment support beneficial effect of FA supplementation for preventing seizure repetition and associated neurodegenerative effects.
