Optimizing the extraction method of collagen biopolymer for the fabricationof skin tissue engineering scaffold

چکیده
Collagen biopolymer, due to its high structural, mechanical and biocompatibility properties, is oneof the most widely used natural polymers in the manufacture of various types of wound dressingsand skin tissue engineering scaffolds. Collagen based scaffold can provide suitable conditions forthe growth and migration of skin tissue cells with the lowest response of the immune system. Inrecent years, the dermis layer of calf skin has been known as a rich source of type Ι collagen for theconstruction of biological scaffolds due to its high abundance, cost-effectiveness, and suitablebiomimicry. Various factors such as solubility, viscosity, thermal stability, synthesis temperature,reducing the degradability of the structure and pre-treatment process will be effective on thequantity and quality of extracted collagen. In this research, an attempt was made to improve thecollagen extraction process by examining the effectiveness of each of these factors: increasing thesynthesis time for ۱ week, increasing the acid volume by two times, reducing the synthesistemperature and decellularizing the tissue. The quantitative percentage of collagen extractedthrough the acid method increased from ۹% to ۵۱% of the primary tissue of the dermis, whichquantitatively became closer to the efficiency of the enzyme extraction process. . The scaffoldprepared in each method was examined by scanning electron microscope in terms of surfacemorphology properties and cross section of the structure. This collagen-based biopolymer scaffoldhas a spongy structure with an average pore size of ۱۵۷ micrometers, suitable for the fabrication ofwound dressings and skin tissue engineering scaffolds.کلیدواژه ها
