Frequency of Quinolone Resistance Associated Genes in Clinical Isolates of E. coli Isolated from Urinary Tract Infections in Rasht

BACKGROUND AND OBJECTIVESFluoroquinolones are a class of broad-spectrum antimicrobial agents which often used for the treatment of lower UTIs. We aimed to determine the frequency of quinolone resistance genes in E. coli isolated from urinary tract infection in Rasht city.MATERIALS AND METHODSResistance of ۲۰۰ clinical isolates of E. coli to common fluoroquinolones and the MIC of ciprofloxacin were determined according to the clinical and laboratory standard institute guideline. Frequency of ۵ plasmid mediated quinolone resistance genes qnrA, qnrB, qnrS were investigated by PCR.RESULTS AND DISCUSSIONAccording to phenotypic assays, ۱۱۰ isolates (۵۵%) were resistant to at least one quinolone compounds and ۵۰ (۲۵%) isolates showed high-level ciprofloxacin resistance. Among the PMQR tested genes, qnrS was the most common PMQR gene detected in this study (𝑛 = ۸۴), followed by qnrB (𝑛 = ۵۶), and qnrA (𝑛 = ۱۷).CONCLUSIONThe potential for dissemination of these resistance genes poses a serious threat to the management of infections by these bacteria and compromised therapeutic options in resistance strains.
