The Expression of MMP۱ and MMP۷ in Mice Liver after Exposure to Aflatoxin B۱ Using Immunohistochemistry Technique

چکیده

Mycotoxin is a class of poisonous secondary metabolites generated by filamentous fungi and found in agricultural commodities worldwide. Therefore, the current study aimed to investigate how aflatoxin B۱ affected hepatic cellular architecture and Matrix metalloproteinase expression in particular (MMP۱ and MMP۷) in the livers of experimental mice (IHC). A total of sixteen mice (four groups) were studied after being given pure aflatoxin B۱ (۹mg/kg B.W., ۶mg/kg B.W., and ۳mg/kg B.W.) (produced from Aspergillus flavus) or a control group (not treated). MMP۱ and MMP۷ expressions were also measured using the MMP۱ and MMP۷ expression assays (IHC). The degree of liver damage is related to the AFB۱ concentration and the duration of exposure. IHC reveals a considerable rise in MMP۱ and MMP۷ expression in the livers of mice given a maximum concentration of ۹۰% ((۹ mg/B.W.) pure AFB۱), which approached the toxin's effect toxic dosage. MMP۱ and MMP۷ expression were also increased by AFB۱ at dosages of ۶۰ and ۳۰% (۶mg/BW and ۳mg/B.W., respectively), although not to the same extent as ۹۰%. MMP۱ was significantly more expressed than MMP۷ compared to control, and AFB۱ at ۹۰, ۶۰, and ۳۰% concentrations caused changes in hepatic cellular architecture, organization, and liver tissue damage and dramatically increased MMP۱ and MMP۷ production in hepatic tissue following treatment. Increased levels of pure aflatoxin B۱ will harm liver tissue and MMP۱ and MMP۷ expression. MMP۱ was more substantially expressed than MMP۷.
