Drug discovery for hub genes based onbioinformatics in Non-Small Cell Lung Cancer

چکیده مقاله:
Background: Lung cancer is the ۲nd most common cancerworldwide. Therefore, according to the importance of this disease,finding the genes and cell pathways involved in this diseaseand finding treatment opportunities has led us to find theseeffective genes using bioinformatics and find the right drugsfor them.Materials and Methods: The bioinformatics analysis of our researchmostly involved the Gene Expression Omnibus (GEO).First, we screened DEGs based on the Transcriptome AnalysisConsole (TAC). Next, the protein-protein interaction (PPI) networkof the DEGs was built in the STRING database. Then,hub genes were screened through the cytoscape. Additionally,using the PHARMGKB we found drugs for our hub genes.Results: We ended up with ۴۳۸۰ DEGs, including ۱۶۳۷ upregulatedand ۲۷۴۳ downregulated DEGs. From the PPI, wefiltered out ten hub genes, and these genes were significantlyupregulated in NSCLC samples, findings consistent with theexpression validation Results: Additionally, survival analysisshowed that high-level gene expression of NEIL۳, GMPS,RFC۴, CDK۱, EXO۱, BRCA۱, PSMD۱۴, MAD۲L۱, CDC۶,and CCNB۱ were connected with the poor overall survival ofNSCLC patients. Using PHARMGKB, we also found candidatedrugs for EXO, BRCA۱, and PSMD۱۴.Conclusion: The present study screened out the essential genesand pathways related to NSCLC pathogenesis, which couldprovide new insights for the future molecular targeted therapyand prognosis evaluation of NSCLC. Based on the informationwe obtained, no other drugs have been proposed to prevent theoverexpression of genes. This study shows that we can finddrugs to prevent the overexpression of other genes.
