A Genome-Wide Association Study on ExcessedBMI and Cancer Risk in Iranian Population

چکیده

Introduction: Body-mass index (BMI) is a hallmark of adiposity,which is a risk factor for many common cancers (۱). Thegenetic architecture of BMI among the Iranian population ispoorly studied. Therefore, we performed a genome-wide associationstudy (GWAS) to detect genetic regions and candidategenes associated with BMI in the so-called population (۲).Materials and Methods: DNA was extracted from ۲۰۰ saliva samples and genotyped using the Infinium Global ScreeningArray, which contains more than ۷۰۰۰۰۰ single nucleotidepolymorphisms (SNPs) with known positions in the human genome.A total of ۳۱۳۱۲۲ SNPs passed the quality control proceduresand allele frequency limitations. Phenotype informationwas collected with questionnaire from ۲۰۰ adults with agesbetween ۱۹ and ۶۵ years, and sixteen individuals did not havedata on BMI and were excluded. The final dataset comprised of۱۹۰۱۱۰ SNPs in ۱۴۹ (۷۴ male and ۷۵ female) participants. Inorder to perform association analysis, a linear regression modelwas used. A p-value < ۰.۰۰۰۱ was considered to be genomewidesignificant. In order to identify associations of genotypeswith BMI, a GWAS were performed.Results: Results from the linear model indicated that rs۱۹۴۹۰۵۲۱۱ ۱۰۸۹۳۲۶۶۲ G (P = ۶.۶۲۱۲۷۶e-۰۶) and rs۵۸۵۰۸۲۷۳ ۱۷۳۱۴۲۲۴۸۴ A (P = ۱.۳۴۸۹۲۴e-۰۵), showed the strongest associationsfor maximum BMI in Iranian population.Conclusion: In this study, we conducted a GWAS of BMI on۱۴۹ individuals from Iranian population. Among ۳۱۳۱۲۲ testedSNPs, rs۱۹۴۹۰۵۲ and rs۵۸۵۰۸۲۷۳ were most associated withhigh BMI. Many studies have shown that increased BMI is associatedwith cancer risk (۳). Hence obesity is major cancer riskfactor (۴), investigations on rs۱۹۴۹۰۵۲ and rs۵۸۵۰۸۲۷۳ for cancerprevention studies is recommended. Further analysis is necessaryto determine more SNPs in other populations, becausegenetic susceptibility may vary by ethnicity. This study, has thepotential to identify individuals at increased risk of obesity inorder to reduce the adverse health effects.
