Identification of Key Genes in Gastric Cancercoagulation Pathway Using Bioinformatics Analysis

Introduction: Gastric cancer (GC) is one of the digestive system'ssystem'smost common and deadly malignancies. According torecent research, coagulation factors contribute to the Pathogenesisof GC. In this study, we used bioinformatics analysisto identify important genes in GC involved in the coagulationsignaling system.Materials and Methods: First, we analyzed the GSE ۶۳۰۸۹ datasetin order to obtain the genes that were significantly changed inexpression. In parallel, we used the msigdb database to obtainthe set of genes participating in the coagulation pathway. Aftersharing the genes obtained from the microarray analysis andthe genes of the coagulation pathway, we compared the geneswith the genes obtained from the TCGA cohort study . Genesthat passed all these steps were entered into GEPIA, UALCAN,kaplan meier plotter and TIMER databases for further studies.Results: Using the MSigDB database, we found ۱۳۸ genes thatplay a role in coagulation pathways. In the next step and afteranalyzing the GSE۶۳۰۸۹ dataset, we filtered the genes consideringadjusted p value <۰.۰۱ and |Log FC| >۲. Next, we sharedthe genes obtained from the previous two filters with the genesthat were significantly obtained from the cohort study fromthe TCGA database. Among the obtained genes, we separatedthose that were significantly changed between all four stages ofgastric cancer. These genes were C۳, CD۹, FGA, FGG, FN۱,MMP۹, SERPINE۱, SPARC and TIMP۱.Conclusion: In this study, we introduced genes that have beenconfirmed by two valuable techniques, RNA seq and microarray,and can be used as new biomarkers in the diagnosis andprognosis of gastric cancer
