The use of mesoporous silica nanoparticles as a gene delivery system was gained attention due to the non-viral and non-organic vectors that offer several advantages, such as reduced toxicity and immunogenicity. The surface engineering of the nano particles may improve not only the biocompatibility, but also enhanced the uptake ability of the nanoparticles. Methodology: In the present study, we use polyethyleneimine as a cellular uptake enhancer for transfection of pEGFPC۱ plasmids in vitro. evaluated the transfection efficiency and toxicity of the nanoparticles using green fluorescent protein plasmid, pGFPC۱, and cytotoxicity assays. Result and Discussion: The electrophoretic analysis revealed that polyethyleneimine-grafted mesoporous silica nanoparticles protected DNA from both ultrasound damage and nuclease degradation. polyethyleneimine-grafted mesoporous silica nanoparticles significantly improved GFP gene expression in HEK ۲۹۳ cells with an acceptable level of viability, and higher transfection efficacy compared to free polyethyleneimine. The flow cytometry results revealed that polyethyleneimine-grafted mesoporous silica nanoparticles improved gene delivery efficiency by over two-fold compared to control. Conclusion:These findings suggest that polyethyleneimine-grafted mesoporous silica nanoparticles have potential as a gene delivery system to improve their efficacy and safety
