Effects of Methylglyoxal and Aspirin on In Vitro Coagulation and Clot Permeability

چکیده

Backgrounds and Aims: Methylglyoxal (MGO) is an –α, β dicarbonyl aldehyde inevitably produced from triose-phosphate intermediates of glycolysis, and amino acid. Increased MGO in blood leads to alterations in coagulation, clot permeability and thus, atherosclerosis in children with diabetes; however, the precise mechanism is not clear. The present study aimed to compare different concentrations of MGO and aspirin on coagulation and clot permeability in the plasma of healthy individuals in vitro. Materials and Methods: Different concentrations of MGO (۵, ۵۰, ۱۰۰, ۵۰۰ µM) and aspirin (۱, ۱۰, ۱۰۰ mg/l) were added to the plasma citrate.  They were incubated at ۳۷°C for ۲۴ h. Then, coagulation parameters were analyzed by the turbidimetric procedure and then clot permeability was investigated. Results: MGO at ۵۰۰ µM with aspirin ۱۰۰ mgl/ made significant changes in the coagulation maximum velocity (۰.۲۵۳±۰.۰۰۶), total coagulation time (۸۰۳±۸.۸۸s) and permeation coefficient (۰.۷۷۸×۱۰-۶±۰.۰۹۹) compared to MGO at ۵۰۰ µM (۰.۲۷۱±۰.۰۰۷), and (۵۰۰±۱۰.۰۰), (۰.۴۴۶×۱۰-۶±۰.۰۱۷), respectively (P< ۰.۰۵). MGO at ۵۰۰ µM with aspirin ۱ mg/l did not significantly change in either parameter (p> ۰.۰۵). MGO at ۱۰۰ µM with aspirin ۱ mg/l did not significantly change in either parameter (p> ۰.۰۵), compared to MGO at ۱۰۰ µM. MGO at ۵ µM with aspirin (۱, ۱۰, ۱۰۰ mg/l) changed in all coagulation and clot permeability parameters (p< ۰.۰۵), compared to MGO at ۵ µM. Conclusions: Our findings revealed that aspirin (≥۱ mg/l) was held to have more effects on higher concentrations of MGO. Moreover, it decreased the velocity of coagulation and increased permeability of clot.
