Vitamin D Binding Protein Gene Polymorphisms and its Association with Type ۲ Diabetes Mellitus in an Iranian Population

Polymorphisms of Vitamin D-binding protein (DBP) may represent a risk factor for susceptibility to Type ۲ Diabetes Mellitus (T۲DM). Two polymorphisms are common at codons ۴۲۰ (ACG to AAG) and ۴۱۶ (GAT to GAG) in the DBP gene. The present study aimed to assess variants of DBP at codons ۴۲۰ and ۴۱۶ utilizing polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP), which were digested by Sty I (codon ۴۲۰) and Hae III (codon ۴۱۶) restriction enzyme. For this purpose, ۲۴۰ patients were recruited along with ۱۵۹ controls. The genotype frequency of Glu/Glu at codon ۴۱۶ in control and patient groups was ۲۵.۱۶% and ۳۲.۹۲% respectively. Moreover, the genotype frequency of Lys/Lys was ۶.۶۷% in patients and ۱.۸۹% in controls at codon ۴۲۰. A significant difference was found between control and patient groups in genotype frequencies at codon ۴۲۰ (p<۰.۰۵). It was found that there might be an association between the vitamin D-binding protein gene and T۲DM.
