چکیده
Objective: The ۶-O chitosan sulfate (CS) with naked aminegroups was synthesized via regioselective modification ofchitosan (C) during which both crosslinking capacity and antibacterialproperties of the C were remained intact. This wasachieved by sulfation of the C under controlled acidic conditionsusing a chlorosulfonic acid/sulfuric acid mixture. Subsequently,a chemically crosslinked hydrogel of the CS was usedas a wound dressing subs trate.Materials and Methods: The modified sulfate groups retainedthe biocompatibility of C and showed antibacterial effectsagains t gram-positive and gram-negative bacteria. In addition,the presence of sulfate groups in the CS chemical s tructure improvedits anticoagulant activity compared to the unmodified C.Results: Both in vitro and in vivo enzyme-linked immunosorbentassay (ELISA) measurements showed that CS had a higherpotential to bind and scavenger anti-inflammatory cytokines,including IL-۶ and transforming growth factor-β (TGF-β), bothof which play critical roles in the early s tage of the wound healingprocess. After treatment of full-thickness wounds with CShydrogels, the macrophage cells (c.a. ۶ × ۱۰۴ cells) expressedsignificantly more M۲ phenotype markers compared to the Cgroup (۴.۲ × ۱۰۴ cells).Conclusion: Furthermore, the CS hydrogel induced better reepithelializationand vascularization of full-thickness woundsin mice compared to the C hydrogel during ۳۰ days.کلیدواژه ها
