چکیده

Objective: In the novel technologies based on tissue engineeringfor regenerative medicine, determining the s tem cells' fateis essential. In this regard, non-invasive tracking of s tem cellsusing natural-based nanos tructures would be a promising approach.Materials and Methods: In this s tudy, chitosan-alginate nanoparticlescontaining Gd۳+ were synthesized using the pregelationmethod for mesenchymal s tem cell tracking usingthe MRI technique. Characterization methods of SEM, FTIR,TEM, and DLS were utilized for nanoprobe evaluations ands tem cell tracking was visualized using an in vivo s tudy. For invivo analysis, ۵ × ۱۰۷ labeled cells per pellet with a volume of۰.۱ mL (subcutaneous) were injected into the dorsal flank of amale BALB/c mouse.Results: Results demons trate that the spherical shape of nanoparticleswith the approximate size of ۸۰ nm was synthesized.ICP result shows the Gd۳+ concentration of۱۹۵۸.۳۳ ppm. The values of r۱ and r۲ in the phantom were۱۲.۸ and ۱۸.۷۵۴ mM−۱s−۱ and the r۱ and r۲ values in cell۷.۵۶ and ۲۲.۱۶۲ mM−۱s−۱ were reported. The r۱ relaxivityfor the nanoparticles in the phantom was ۱۲.۸ mM−۱s−۱ perGd۳+ ion, which is ۳.۵ times larger than that for Dotarem®(r۱ -۳.۶ mM−۱s−۱ per Gd۳+ ion) and the r۱ relaxivity for thenanoparticles in s tem cells was ۷.۵۶ mM−۱s−۱ per Gd۳+ ion,which is ۲.۱۶ times larger than that for Doratem®. The significantlysignal enhancement in t۲-weighted in in vivo images wasobserved in labeled nanoparticles.Conclusion: Our results reveal that the aforementioned biocompatiblenanoparticles have a high potential for mesenchymals tem cell tracking for tissue engineering applications.
