چکیده

Objective: Neuronal cell death and axonal degeneration arecommon pathophysiological occurrences after spinal cord injury(SCI). Improving s tem cell differentiation to replace los tand damaged cells after SCI might be a potential approach.Many inves tigations are now underway to es tablish proceduresfor motor neuron differentiation in order to regenerate cells thatare similar to the los t endogenous ones as a potential cure forSCI and neurodegenerative illnesses. The goal of this work wasto see how epothilone B (EpoB) affected the differentiation ofhuman endometrial s tem cells (hEnSCs) into motor neuron(MN) like cells in the presence of neurite outgrowth inhibitorA (Nogo-A).Materials and Methods: hEnSCs were differentiated into motorneuron‐like cells in three s teps (pre‐induction, induction,and maturation). The Epo B differentiation group was treated with ۱۰ nM EpoB (added in s teps ۲ and ۳). The cells were differentiatedin the presence of NogoA (۴۰۰ ng/mL) to simulatethe in vivo inhibitory environment. Cell alignment and neuriteelongation were observed and measured using an invertedphase-contras t microscope. To reveal the morphology of cells,they were s tained for β‐III‐tubulin marker by immunofluorescent(IF).Results: The neurite length of the EpoB group significantly (P< ۰.۰۰۱) increased in comparison with the control group. IFimages indicated the s tability of neuronal morphology, greatcell alignment, and neurite elongation in the EpoB group in thepresence of Nogo‐A.Conclusion: Despite the presence of the Nogo‐A inhibitor, thiss tudy showed EpoB as a boos ter for neurite elongation and anotable cell alignment organizer during MN differentiation
