PIWIL۲ Promotes The Growth and ColonizationRate of Colorectal Cancer Cells

Objective: Unlimited proliferation caused by unbalanced cellcycle progression and apoptosis is responsible for tumor initiation,development and metas tasis. In this research, the role ofPIWIL۲ overexpression on this imbalance was inves tigated inSW۴۸۰ cells as a colorectal cancer cell (CRC) line.Materials and Methods: PIWIL۲-overexpressing and controlSW۴۸۰ cells were es tablished by transfecting PCDNA۳-PIWIL۲ and PCDNA۳-empty vectors into SW۴۸۰ cell line,respectively via electroporation. The relationship between thePIWIL۲ gene overexpression on the rate of proliferation, cellcycle progression and apoptosis genes were determined by realtimepolymerase chain reaction (PCR), MTT assay, DoublingTime assay and colony formation.Results: A significant increase in S tAT۳, BCL۲-L۱, BCL۲-L۲,cyclin D۱ and Ki-۶۷ genes expression were found in SW۴۸۰-PIWIL۲ compared to SW۴۸۰-control cells. Colony formationanalysis showed a ~۴-fold increase in the number of colonies (Pvalue <۰.۰۰۱). Furthermore, the results of Doubling Time assayand MTT indicated that PIWIL۲ overexpression induced timedependenteffects on proliferation rate.Conclusion: In this s tudy our data indicated that PIWIL۲ overexpressionpromoted the proliferative capacity of CRC cells aswell as cell colonization and inhibition of apoptosis in thesecells. These data revealed the importance of PIWIL۲ gene as avaluable oncogenic biomarker for the purpose of cancer diagnosisand targeted therapies.
