Objective: Mesenchymal s tem cell (MSC) therapy is an amazingrevolution in clinical therapy of various skeletal diseasesuch as bone defects. Using crude herbal extracts such as Shilajitwas common in bone fracture repair. The purpose of the currents tudy was to discover the synergic effects of ASCs (human adipose derived mesenchymal s tem cells) and Shilajit in combinationof alginate on the repair of rat tibia bone defect.Materials and Methods: Seventy-five healthy Sprague Dawley(SD) rats were used in this s tudy. A ۷ mm defect was madein right tibia of each rat, and then they were divided into fivegroups including alginate+Shilajit+ASCs, alginate+ASCs,alginate+Shilajit, alginate, and untreated control groups. Specimenswere collected at ۲ and ۴ weeks after surgery and thenbone formation was assayed via radiology, H&E and trichromeMasson s taining, and biomechanical tes ts.Results: The population doubling time (PDT) tes t indicated asignificant decrease in doubling time of treated ASCs than ofthe untreated group (P value <۰.۰۱). The X-ray photography oftibia specimens showed less repair in the alginate and untreatedgroups than all other groups at weeks ۲ and ۴. In vivo morphometricallyes timations showed predominant synergic effectsof alginate+Shilajit+ASCs on increasing the number of os teoblasts, os teocytes, and bone area relative to the alginate+ASCs,alginate+Shilajit, alginate, and untreated groups at week ۲ (Pvalue <۰.۰۰۰۱). Beside, s trength of rat tibia bone showed significantelevation in the alginate+Shilajit+ASCs compare tothe alginate+ASCs (P value <۰.۰۵), alginate+Shilajit (P value<۰.۰۱), alginate, and untreated groups (P value < ۰.۰۰۱ forboth). Trichrome Masson s taining revealed that collagen depositionwas higher in the alginate+Shilajit+ASCs compare to theother groups at week ۲.Conclusion: The combination of ASCs, Shilajit and alginatecause acceleration and promotion of bone tissue regenerationand provide a suitable approach for cell therapy of bone defects.
