Objective: Os teoporosis (OP) is a chronic progressive metabolicbone disease caused by the reduced bone formation andincreased bone resorption. Recently, applying mesenchymals tem cells (MSCs) has gained a huge amount of interes t amongscientis ts. Hypoxic preconditioning (HP) of MSCs is an efficients trategy of reducing apoptosis of the s tem cells and enhancingtheir regenerative ability and therapeutic potentials forthe treatment of OP. The present s tudy aimed to examine theeffect of injection of human adipose-derived MSCs pre-conditioningwith Cobalt Cholride on the os teogenesis in Wis tarovariectomized rats.Materials and Methods: In this s tudy, os teoporosis was inducedby bilateral ovariectomy. In the pilot s tudy, human adipose-derived MSCs were pre-conditioned with ۱۰۰, ۲۰۰ and۴۰۰ μM cobalt chloride for ۲۴ and ۴۸ h. According to MTT andAcridine Orange Ethidium Bromide s taining results, ۱۰۰ μMcobalt chloride for ۴۸ h was used to continue working. Fourmonths after ovariectomy, rats were then divided into threegroups receiving pre-conditioned hypoxic cells, normoxic cellsand saline buffer phosphate. At ۱۲ and ۱۶ weeks, sampling wasperformed to evaluate the expression of Runx۲, OPG and OCproteins in tibia by wes tern blotting.Results: The results showed that in the group receiving hypoxiccells in both weeks ۱۲ and ۱۶, the expression of Runx۲, OPGand OC proteins was significantly increased compared to thesham and normoxic groups.Conclusion: Concluded that artificially controlled direction ofMSC migration could remodel the micros tructure of bone tissue,which led to the appropriate os teogenesis
