Crucial Role of Meiosis Recombination and DNARepair Genes in Diminished Ovarian Reserve (DOR)

Background: Diminished ovarian reserve (DOR) refers towomen under the age of ۳۸ years, whose number and qualityof the oocyte pool in their ovaries have decreased. DOR doesnot have an obvious phenotype whils t early detection of DORis very useful to increase the chances of successful fertility. Recents tudies have shown that the pathogenesis of DOR is likelyto be multifactorial. Little is known about the molecular andgenetic factors associated with ovarian reserve. DNA doublestranded breaks and failure to repair them seemed to be one ofthe main factors for decrease in ovarian reserve according toreported mutations in pedigree analyses with ovarian reservediseases. The relationship between alteration of genes associatedwith meiosis and homologous recombination with DOR hasnot been s tudied so far. The aim of this s tudy was to evaluatethe expression level of MSH۴, PSMC۳IP, HFM۱, SYCE۱ andMCM۹ genes, involved in meiosis and homologous recombination,in granulosa cells of women with DOR.Materials and Methods: Granulosa cells were isolated fromfollicular fluid of ۱۰ women with DOR and ۱۰ healthy controlwomen, referred for male factor infertility, with minimumnumber of epithelial cells. RNA extraction and CDNA synthesiswere performed. The expression level of the genes was assessedby using q-RT PCR. p value <۰.۰۵ was considered ass tatis tically significant differences.Results: The expression level of MSH۴, PSMC۳IP and SYCE۱genes in women with DOR compared to controls greatly decreased(P<۰.۰۰۱), additionally MCM۹ gene expression inDOR significantly decreased (P=۰.۰۰۹). Although HFM۱ genealso decreased in the DOR group but it was not s tatis tically significant(P=۰.۱۱۲).Conclusion: Due to significant reduction in the expression levelof genes involved in meiosis recombination and DNA repairincluding MSH۴, PSMC۳IP, SYCE۱ and MCM۹, these pathwaysseemed to have a crucial role in quantity and quality ofoocyte pool certainly in DOR.
