Background: Diminished ovarian reserve (DOR) is a commondisease that is identified by a decrease in ovarian reserve fromnormal range. DOR is a multi-factorial disease that affects ۱۰%of women seeking fertility treatment. Although DOR is common,its etiology is less well known. S tudies show that genesinvolved in the meiotic recombination pathway are associatedwith ovarian reserve; among them, we selected S tAG۳, MCM۸,DMC۱, FANCM, RBBP۸ and MSH۵ genes which only theirmutations had previously been reported in ovarian reserve diseases.The goal of this s tudy was to evaluate the expression ofgenes involved in meiosis in granulosa cells of women withDOR.Materials and Methods: Follicular fluid was collected from۱۰ women with DOR and ۱۰ healthy women as control group,referred for male infertility factors, and isolating of granulosacells with minimum epithelial cells was processed. RNA extraction,cDNA synthesis and RT-qPCR were performed. Geneexpression was compared between DOR and control groupsand P value<۰.۰۵ was considered as a significant difference.Results: Expression of S tAG۳, MCM۸, DMC۱ and FANCM genes greatly decreased (P<۰۰۱) and RBBP۸ gene downregulated(P<۰۰۷) in women with DOR. Although MSH۵ gene expressionin DOR was lower than controls, it was not s tatis ticallysignificant (P<۰.۰۷۸).Conclusion: According to downregulation of S tAG۳, MCM۸,DMC۱, FANCM and RBBP۸ genes, the relationship betweenmeiotic genes alteration with ovarian reserve seemed to be crucial.These data are useful to better unders tand DOR etiologyfor providing more appropriate medical services
