Background: Polycys tic ovary syndrome (PCOS) is one ofthe mos t common endocrine disorders in women of reproductiveage. The pathophysiology of PCOS is complex, therefore,in the present s tudy, we simultaneously inves tigated Salviaofficinalis extract as an antioxidant and vitamin E in PCOSmice.Materials and Methods: In this experimental s tudy, PCOmodel was developed in adult female mice during ۸ weeks.The ۳۰ mice were divided in five groups include control, PCOmodel (using es tradiol valorite or EV at ۴۰ mg/kg), Salvia officinalisextract (۴۰۰ mg/kg day), vitamin E (۶۰ mg/kg/day,orally), and the Salvia officinalis extract + vitamin E model.After mice sacrifing, their ovarian tissue was removed andhis tological and morphometric s tudies were performed. Aftertissue lysis, total antioxidant capacity was measured by FRAPmethod and oxidative s tress was measured by flow cytometry.Results: The results of tissue sections showed that EV administration caused cys t formation in the s tudy groups. Cys t formationand the percentage of preantral and antral follicles inthe group that received Salvia officinalis extract and vitamin Etogether were significantly lower (P≤۰.۰۰۱) than the groups thatreceived each alone. In the group that received a combination ofboth antioxidants and vitamin, the level of oxidative s tress andtotal antioxidant capacity were the lowes t and highes t, respectively,compared to the other groups.Conclusion: The results showed that co-adminis tration of Salviaofficinalis extract and vitamin E in PCO mice, in addition toreducing the amount of oxidative s tress by increasing the antioxidantproperties, but also by reducing the number of cys ts andimproved the symptoms of this syndrome
