چکیده

Background: Chronic and high dose consumption of nanomicellecurcumin (NMC) has been shown to negatively affectthe tes ticular antioxidant s tatus, spermatogenesis development,sperm quality, and pre-implantation embryo development. Thecurrent s tudy tried to uncover a new aspect of NMC-inducedimpact in the tes ticular tissue by more focusing on lactate andglucose transportation in Sertoli-germ cell network.Materials and Methods: For this purpose, ۲۴ mature maleWis tar rats were divided into control (normal saline, n=۶) andNCM-received groups (n=۱۸). The NMC group subdivided into۷.۵ mg/kg, ۱۵ mg/kg, and ۳۰ mg/kg NCM-received rats (n=۶ rat/group, orally). Following ۴۸ days, the tes ticles were dissectedout, the mRNA levels of monocarboxylate transporter (MCT-۴,a lactate transporter on Sertoli cells), and glucose transporters ۱and ۲ (GLUTs ۱, ۲) as well as GLUT+ cells (Sertoli and germ)dis tribution in the seminiferous tubules were analyzed andcompared between groups. The data were s tatis tically analyzedby one way ANOVA and continued by bonferroni tes t.Results: Observations showed that NMC, mainly at higherdoses, could significantly (P< ۰.۰۵) decrease MCT-۴, GLUT-۱,and GLUT-۲ mRNA levels in the tes ticular tissue compared tothe control rats. Moreover, the ۱۵ mg/kg and ۳۰ mg/kg NMCreceivedrats revealed a remarkable (P< ۰.۰۵) reduction in theGLUT-۱+ and GLUT-۳+ germ and Sertoli cells per seminiferoustubules with the same criteria.Conclusion: Our data showed that chronic and high-dose consumptionof NMC is able to significantly affect the glucose andlactate transportation in the tes ticular tissue by suppressing theexpression levels of lactate and glucose transporters
