Sperm Medium Supplementation: Effects of L-Prolineon Human Sperm Function In Vitro

Background: Incubating spermatozoa is a routine procedurein infertility centers, so supplementation of sperm culture withantioxidants is recommended to preserve sperm quality.Materials and Methods: Sperm samples were collected fromthe normozoospermic man who received antioxidants (vitaminE and D). Each sample was divided into four portions: increasingconcentrations of L-proline (۰, ۱, ۲, ۴ mmol/L) were addedto the sperm culture. Sperm parameters and chromatin integrity,including Aniline blue (AB), Toluidine blue (TB), ChromomycinA۳ (CMA۳), and Sperm chromatin dispersion (SCD), wereevaluated. The levels of reactive oxygen species (ROS), lipidperoxidation (LPO), and total antioxidant capacity (TAC) weredetermined in the sperm medium. All assessments were performedafter ۰, ۱, ۴, and ۲۴ hours of incubation.Results: ۲ and ۴ mmol/L L-proline significantly improved preservationof motility and viability after ۲۴ hours of incubation,and ۲ mmol/L of L-proline maintained normal morphologycompared to the control group. The level of ROS significantlydiminished in the ۴ mmol/L L-proline group compared to thecontrol group. ۲ and ۴ mmol/L L-proline decreased MDA levelsafter ۴ and ۲۴ hours of incubation. ۲ mmol/L of L-proline increasedmaintenance of TAC levels compared to other groups.Supplementing ۲ and ۴ mmol/L L-proline significantly improvedthe AB, TB, and CMA۳ levels. ۲ mmol/L L-proline significantlyreduced SCD levels compared to the control group.Conclusion: The inclusion of L-proline in the human spermmedium maintains sperm parameters and chromatin qualityduring different incubation times.
