Background: Polycys tic ovary syndrome (PCOS) is related tooxidants-antioxidants imbalance. Previous research has shownthat conditioned media-derived mesenchymal s tem cells play a role in improving antioxidant capacity in the different cellcultures. Therefore, we inves tigated the effects of conditionedmedia of human umbilical cord Wharton's jelly-derived mesenchymals tem cells (hWJMSCs-CM) on the expression of antioxidantenzymes of human normal granulosa cells (N-GCs) andgranulosa cells from polycys tic ovaries (PCO-GCs).Materials and Methods: Ovarian GCs were obtained duringthe oocyte retrieval procedure from ۱۰ women with PCOS andfrom ۱۰ healthy women who were referred to Shiraz GhadirMother and Child Hospital. To assess superoxide dismutase(SOD) and glutathione peroxidase (GPx) genes expression, totalcellular RNA was extracted from GCs using a total RNAPurification Mini kit. Extracted mRNAs were reversely transcribedinto firs t-s trand cDNA using a cDNA synthesis kit. qRTPCRwas accomplished using SYBR Green PCR Mas ter Mix.Samples were quantified by the comparative ΔΔCt method withβ-actin as an internal control. Experiments were performed intriplicate.Results: Basal SOD and GPx genes expression by N-GCs wassignificantly higher than PCO-GCs. hWJMSCs-CM significantlyincreased SOD and GPx genes expression in N-GCs andPCO-GCs. In the presence of hWJMSCs-CM, GPx genes expressionin the PCO-GCs was not significantly different fromN-GCs.Conclusion: hWJMSCs-CM had positive impacts on luteinizedGCs through its effect on the expression of antioxidantenzymes. It can improve PCO-GCs to normal conditions by affectingthe main aspects of oxidative metabolism.
