چکیده
Background and Aim : Biosurfactants are amphiphilic molecules that are mainly produced bymicroorganisms as extracellular secondary metabolites and are of special importance due to theiruse in various industries. The aim of this research was to isolate and identify Lactobacillus strainsproducing biosurfactants from local dairy products and to investigate the antimicrobial effects ofbiosurfactants produced by them.Methods : Dairy products were sampled and cultured in MRS medium. Hemolytic activity,emulsification, and measurement of surface tension were measured and the isolated strains withthe ability to produce biosurfactants were identified based on biochemical and molecular tests.Then the biosurfactant produced by the selected strain was extracted from the culture medium andthe effect of various factors such as Temperature (۲۵, ۵۰, ۷۵, ۱۰۰), pH (۳-۱۲), and NaClconcentration (۵-۲۰%) were investigated on its stability. Finally, the antimicrobial effect of theproduced biosurfactants on Escherichia coli (PTCC ۱۳۳۰), Staphylococcus aureus (PTCC ۱۱۱۲),Staphylococcus epidermidis (PTCC ۱۸۵۶), Proteus mirabilis (PTCC ۱۷۱۰) and Pseudomonasaeruginosa (PTCC ۱۰۷۴) by measuring MIC and MBC was evaluated.Results : In this study, ۴۷ bacterial strains were isolated. Among them, ۱۳ strains had hemolyticactivity, and ۵ strains had emulsifying activity above ۷۰%. Finally, only ۱ strain was able to bringthe surface tension to less than ۴۰ mN/m. Based on biochemical and molecular tests, the isolatedstrain of Lactobacillus plantarum was identified. Also, the results showed that the producedbiosurfactants had antibacterial activity and the most susceptible and resistant bacteria to theproduced biosurfactants were Staphylococcus aureus and Pseudomonas aeruginosa respectively.Conclusion : The isolated Lactobacillus plantarum has a high ability to reduce surface tension andthe biosurfactant extracted from it has good stability and antimicrobial effects. For this reason, itcan be used in various industries.کلیدواژه ها
