Investigation of allergenicity of antimicrobial peptidesMagainin and MSI-۹۹ using in silico method

Background and Aim : Due to the increasing use of antibiotics and the increasing resistance ofmicroorganisms to them, it will be important to study to find alternative compounds for antibiotics.Antimicrobial peptides can be used as an alternative to antibiotics. Magainin peptide and its analogMSI-۹۹ contain ۲۳ amino acids and have a cationic and hydrophobic structure. These peptidescause damage to the cell membrane by creating alpha-helical ion channels. The purpose of thisstudy is to investigate the allergenicity of these peptides using the In silico method.Methods : In this study, the MSI-۹۹ and Magainin protein sequences were received from the genebank. The complete sequences of these peptides were then searched in the Allergen database at"http://www.Allergenonline.com" and the Allermach database at "http://www.allermatch.org". Inthis way, the complete sequences and the sequences of six to eight amino acids were comparedwith the allergen sequences in the database, respectively. Then, the sequences of these peptideswere enzymatically digested with pepsin and trypsin enzymes in silico, and the resulting fragmentswere examined in terms of allergenicity.Results : The results of MSI-۹۹ antimicrobial peptide homology study with different methods didnot reveal any allergenicity. The fragments obtained from the enzymatic digestion of MSI-۹۹ alsohad no match with the allergen peptides in the database.Conclusion : According to the rules of WHO and FAO regarding complete sequences and shortpeptide sequences, a similarity of more than ۵۵% and ۹۵% with allergen sequences can besignificant. Our results proved that Magainin and MSI-۹۹ peptides are not allergenic. Therefore,this research can solve the concerns related to the use of these peptides as a suitable alternative forantibiotics.
