چکیده مقاله:

Background and Aim : Probiotics, live cells with different beneficiary characteristics, have beenextensivelly studied and explored commercially in many different products in the world. Theirbenefits to human and animal health have been proven in many researchs. Consumer demands forfoods promoting health while preventing diseases have led to development of functional foods thatcontain probiotic bacteria. Lactic acid bacteria (LAB) members, Lactobacilli as normal flora ofgastrointestinal tract have beneficial effects on human health. The aim of this study is to Isolateand identify probiotic bacteria of lactobacillus strains from dairy sewage.Methods : waste water samples were collected from a Tehran Dairy Co in accordance withstandard procedures and enriched in Man-Rogosa-Sharpe (MRS) broth and isolated by growingon MRS agar medium. Small white grown colonies with a semi mucoid consistency were isolatedon MRS medium. Gram-positive, catalase-negative, and oxidase-negative bacilli and coccobacilliwere selected and subcultured on the new MRS agar culture medium. On new subculture,biochemical tests of sugar fermentation , CO۲ gas production , acidic conditions (pH: ۲,۳,۴) andbile salts (۰.۳), was assessed to investigate probiotic characteristics.Results : Our findings demonstrated that the identified Lactobacillus spp has a suitable resistancein pH=۴ (۷×۱۰۶ CFU/mL). In addition, The highest tolerance against to bile salts is (۵×۱۰۵CFU/mL after ۱h ).Conclusion : According to the results of the present study, dairy sewage is a highly potential anda cheap source for isolating Lactobacillus spps and It also give us a wider perspective to make newprobiotic products.
