چکیده مقاله:

Background and Aim : The increasing prevalence of the COVID-۱۹ disease has led scientistsaround the world to look for differences in susceptibility to COVID-۱۹ in different populations.Research has shown that genetic differences in individuals are a factor influencing the functioningof the host immune system when dealing with various infectious agents, including viruses. Sincethe presence of SNPs in genes involved in innate human immune system is influential, weinvestigated the association between host genetic polymorphism in TLR۳ gene and its associationwith COVID-۱۹ disease in a sample of Iranians.Methods : In this study, ۲۳۵ and ۲۱۸ patients with COVID-۱۹ Which had a positive moleculartest result considred as case group for genotyping TLR۳ rs۳۷۷۵۲۹۱ and TLR۳ rs۳۷۷۵۲۹۶respectively. also ۱۰۶ and ۹۸ suspected patients to COVID-۱۹ disease Which had a negativemolecular test result considred as control group for genotyping TLR۳ rs۳۷۷۵۲۹۱ and TLR۳rs۳۷۷۵۲۹۶ respectively.Studied population selected from Ghods clinic affiliated to TehranMedical Sciences, Islamic Azad University, Tehran. Genomic DNA was extracted fromnasopharyngeal and oropharyngeal specimens and genotyping of gene polymorphisms wereperformed using Polymerase Chain Reaction Restriction Fragment Length Polymorphism (PCRRFLP)method.Results : In TLR۳ rs۳۷۷۵۲۹۶, frequency of CC, CA and AA genotypes was ۷۳.۴%, ۲۴.۸% and۱.۸% in patints group and ۸۳.۷%, ۱۴.۳% and ۲% in controls group, respectively.In TLR۳rs۳۷۷۵۲۹۱,frequency of CC, CT and TT genotypes was ۵۵.۴%, ۳۷.۴% and ۷.۲% in patints group,respectively. Also in controls group were ۶۴.۲%, ۳۰.۲% and ۵.۶%. The frequency distributions ofCC, CT, TT TLR۳ rs۳۷۷۵۲۹۱ and AA, CC TLR۳ rs۳۷۷۵۲۹۶ genotypes were not significantlydifferent between those with COVID-۱۹ and suspected PCR-negative individuals. Only TLR۳rs۳۷۷۵۲۹۶ CA genotype frequency (P = ۰.۰۲۱, OR = ۲.۷۰۳, ۹۵% CI: ۱.۱۶۱-۶.۲۹۷), especially inmales between case and control groups, had a significant relationship with susceptibility toCOVID-۱۹.Conclusion : According to the results of this study, polymorphism in TLR ۳ rs۳۷۷۵۲۹۶ can beconsidered in males as one of the factors that increase the susceptibility to COVID-۱۹ and mayalso affect the progression of the disease.
